"Supplemental Figure 1. Lysates (15 µg/lane) of quiescent sperm were separated by PAGE under non-reducing (left lane) or reducing (right lane) conditions and immunoblotted. The antibody to HK1 (H-95) detected the dimer band (arrow) in the non-reducing condition (-&bgr;-mercaptoethanol; BME) and monomer band in the reducing condition (+ BME).
Supplemental Figure 2. Lysates and pellets of sperm activated by incubating in M2 medium for 5 min (- capacitation) and of sperm incubated under capacitating conditions for 2 h (+ capacitation). Lysates and pellets from 0.5 × 107 sperm were separated by PAGE under reducing conditions and immunoblotted with antibodies to the SSR domain of HK1S, P-Tyr, and &agr;-tubulin (loading control).
