"Control images for immunocytochemistry in JAr cells (A-C) and CT-1 cells (D-F). JAr cells were probed with fluorescein (Alexa Fluor (AF) 488) labeled donkey anti-sheep (A), goat anti-rabbit (AF 568) (B), and DAPI (C). CT-1 cells were probed with fluorescein (AF 488)-labeled goat anti-mouse (D), goat anti-rabbit (AF 568) (E) and TO-PRO-3 iodide (F). All the experimental conditions, i.e. cell fixation, reagent concentrations, incubation times and image capture standards, were identical to the experimental conditions employed in Figure 6. "Association of endogenous ETS2 with the regulatory region actively transcribed IFNT from bovine CT-1 cells. Sheared chromatin prepared from formaldehyde-fixed CT-1 cells was analyzed by ChIP. Chromatin was exposed to no antibody (lane 1), anti-DLX3 (lane 2), anti-ETS2 (lane 3), purified rabbit IgG (lane 4), and anti-RNA polymerase II (lane 5). The DNA recovered from each immunocomplex was amplified with primers specific for a region (-188 to +3) containing the ETS2/DLX3 enhancer region within the IFNT promoter. Lane 6 represents the PCR product amplified from 10% of the total input chromatin. Lane 7 shows the PCR product amplified from 0.6 ng of plasmid DNA containing the -457luc reporter construct."
